Citrulline, also known as aminoformylornithine in Chinese, has the chemical name L-Citrulline. Its chemical formula is CHNO, with a molecular weight of 175.19 and the CAS number 372-75-8. The pure form is a white columnar crystal with a melting point of 235.5ºC. It is soluble in water but insoluble in ethanol and ether. In 1914, Japanese scholar Katsuto Okada first isolated it from watermelons and melons, and it was later confirmed by Katsutoshi Watanabe [2] [11] [29].
This substance is mainly found in the fruits of cucurbit crops such as watermelons and melons, as well as the wood of Japanese birch trees. The content of citrulline in watermelons ranks first among cucurbit crops, with yellow fleshed watermelons having a higher content than red fleshed ones [1] [10] [24]. It is synthesized in the urea cycle from ornithine and carbamoyl phosphate, and since the ASS and ASL enzymes that catalyze the decomposition have low activity in the intestine, it is released directly into the blood without decomposition. It can also be a by-product of arginine converting to nitric oxide [4-5] [14]. Production methods include extraction and separation, chemical synthesis, fermentation, and microbial enzyme methods. Among them, the microbial enzyme method has a high product purity and no D-type optical isomers [2] [7] [11] [13]. Detection methods mainly include high-performance liquid chromatography and spectrophotometry [3] [20]. The normal value in serum is 20-40 μmol/L. An abnormal increase suggests citrulline hyperemia [6] [27-28].
It is used clinically for the diagnosis of rheumatoid arthritis and the assessment of sepsis treatment. Its pharmacological effects cover antioxidant defense (removing hydroxyl free radicals), increasing arginine levels to promote NO synthesis, enhancing immunity, delaying aging, and improving heart and lung function. It has an improving effect on male sexual dysfunction and radiation-induced intestinal damage, and is widely used in the fields of medicine, health care, and food.
