NAD
NAD (nicotinamide adenine dinucleotide), also known as coenzyme I, is formed by the connection of nicotinamide, adenine, and two riboses through a phosphodiester bond. It exists in both oxidized form (NAD) and reduced form (NADH), serving as an electron carrier in various enzymatic reactions, achieving oxidation-reduction functions through the transfer of hydrogen anions (H-) and protons (H+). NAD plays a crucial physiological role in the body, participating in energy metabolism processes such as alcohol metabolism, glycolysis, fatty acid oxidation, and the tricarboxylic acid cycle. It is also a co-substrate for enzymes such as Sirtuins, PARP, CD38/157, and SARM1, and can act as a NAD cap to regulate the stability and translation level of mRNA [5] [6]. The NAD synthesis pathway includes the salvage pathway, the Preiss-Handler pathway, and the de novo synthesis pathway. The precursors for synthesis include vitamin B3 and its derivatives (nicotinamide, nicotinamide riboside) and amino acids (aspartic acid, tryptophan, aspartic acid) [5] [7] [8]. NAD-consuming enzymes (including Sirtuins, PARP, CD38/157, and SARM1) consume NAD when exerting regulatory effects, and the decomposed product NAM is utilized by the NAD salvage synthesis pathway to regenerate NAD and maintain stable cellular NAD levels [7]. A decrease in NAD levels is a hallmark of aging and various related diseases, including neurodegenerative diseases (such as Alzheimer's disease, Parkinson's disease, and amyotrophic lateral sclerosis), liver diseases (such as non-alcoholic fatty liver disease), kidney diseases (such as acute kidney injury, chronic kidney disease), blood system diseases (such as hemorrhagic shock), muscle diseases (such as mitochondrial myopathy, muscular dystrophy), and cardiovascular system diseases (such as coronary artery disease, myocardial infarction) [5]. Therefore, stimulating NAD synthesis or inhibiting excessive NAD consumption becomes a potential therapeutic strategy for related diseases. Studying the metabolism and function of NAD is of great significance for understanding disease mechanisms and developing anti-aging therapies.

Nicotinamide Adenine Dinucleotide Nicotinamide Supplement or Health and Antioxidant1.Energy Metabolism
NAD participates in the breakdown of sugars, fats, and proteins in mitochondria, converting them into ATP (the currency of cellular energy). A deficiency of NAD can lead to reduced metabolic efficiency, causing fatigue and decline in organ function.
2.DNA Repair
By activating PARPs (poly(ADP-ribose) polymerase), NAD helps repair DNA damage and maintain genomic stability. Weak DNA repair capacity is closely associated with cancer and age-related diseases.
3.Delaying Aging
NAD is an essential cofactor for the Sirtuins protein family. These proteins are involved in regulating cell lifespan, antioxidation, and metabolic balance. Animal experiments have shown that increasing NAD levels can delay aging and improve metabolic syndrome.
